・A2ARを介したアデノシンシグナル伝達により、様々な免疫抑制作用が生じ、腫瘍の増殖を促進させる。
・AZD4635はA2AR受容体に選択性の高い拮抗作用を持つ経口薬であり、アデノシンによるT細胞抑制作用を元に戻すことが示された。
・AZD4635単剤及び抗PD-L1抗体との併用療法は、高アデノシン及び低アデノシンを示す同系腫瘍モデルにおいて腫瘍増殖を著しく減少させた。
・こうした作用は免疫不全動物では起こらず、免疫介在性作用機序が確認された。AZD4635による標的分子への作用(ターゲットエンゲージメント)については引き続き研究を進めていく。
・これらのデータは、AZD4635が、単剤及び他のガン免疫療法との併用療法により腫瘍のアデノシンレベルとは無関係に、免疫応答性を回復させ、抗腫瘍効果をもたらす可能性があることを示唆している。
・なお、固形がん患者を対象に、単剤治療及びアストラゼネカ社の抗PD-L1抗体であるIMFINZI(durvalumab)との併用療法として、AZD4635の第I相臨床試験を現在実施中です(NCT02740985）。

"Bio Century (Reprint From 2018/3/26)"：「医薬品開発：A2A拮抗薬の将来性」
・これまで専ら神経領域のターゲットとして考えられてきたアデノシン経路が癌免疫領域において興味を持たれている。その理由は、免疫細胞を活性化または抑制させるように制御できるためである。
・当初神経疾患に対して開発されてきたA2A受容体拮抗薬に関する初期の成績については玉石混交であった。しかし、有効性を高めることを目指した新しいアプローチに粘り強く取り組んでいる製薬会社が数社存在する。
・そーせいグループの執行役副社長、チーフR&Dオフィサーであるマルコム・ウィアー氏は、
・BioCentury誌に、A2A受容体が免疫細胞に存在することは十年以上前から知られていたが「この機序の臨床的意義に対する期待が高まったのはチェックポイント阻害薬が登場してからのことである」と述べた。
・アデノシンがアデノシンA2A受容体（ADORA2A）に結合すると、CD8陽性T細胞やその他のリンパ球による炎症性サイトカインの産生を減少させ、免疫抑制性サイトカインの産生を増加させる。
・A2A受容体が活性化することで、PD-1などT細胞の免疫チェックポイントのアップレギュレーションを起こす。
・これによりCD4陽性T細胞におけるフォークヘッドボックスP3（FOXP3）遺伝子の発現が上昇することで、この細胞が免疫抑制機能を持つ制御性T細胞の表現型に分化する。
・そーせいグループを含む最低5社ががんを対象としたA2A受容体阻害薬の開発を行っている。
・そーせい社が提携しているアストラゼネカ社は、バイオテクノロジーで開発したA2A拮抗薬のHTL001071（AZD4635）と抗CD73モノクローナル抗体のMEDI9447の併用療法の後期第I相／第II相試験を、
・EGFR遺伝子変異陽性の進行NSCLC患者98名を対象に臨床試験を行っている。ClinicalTrials.govによるとデータは2021年になる見込みである。

本年度の巨額赤字も決算発表前の下げ考えるとこの段階で織り込んだと考えてもよさそうやね。（Minaの増資ガー。。プロレスはやる可能性あるけど）
