・RNA therapeutics specialist MiNA Therapeutics plc has secured £23m in a Series A financing to push development its first-in-class small activating RNA (saRNA) candidates.
Mina Therapeutics plc’s Series A round was led by aMoon, Israel’s largest healthtech and life sciences venture fund, with participation from existing investors Sosei, Golden Triangle Partners and Galloway.
The cancer therapy specialist said it will use the assets to advance and expand its pipeline of saRNAs,
designed to harness gene activation mechanisms to fight cancer, fibrosis etc., particularly Mina Therapeutics' Phase I hepatocellular carcinoma candidate MTL-CEBPA.
Clincal Phase Ib data on MTL-CEBPA therapy, in combination with sorafenib presented at ASCO conference in June,
suggest that an increase of the C/EBP-alpha master regulator levels reduced the amount of immunosuppressive myeloid cells in the tumour microenvirenment and thus improves the efficacy of sorafenib.
Though the drug combo was generally well tolerated, it caused 15 SAEs in 10 of 37 patients who were enrolled in the study.
saRNA medicines have shown potential to transform the therapeutic landscape of cancer, chronic metabolic and genetic diseases.
The funds will enable MiNA Therapeutics to conduct a Phase II study of MTL-CEBPA in combination with sorafenib in patients with advanced liver cancer and TIMEPOINT,
an ongoing Phase I/Ib study of MTL-CEBPA in combination with pembrolizumab in patients with advanced solid tumour malignancies.

Seven years after leaving investment banking to take the helm at MiNA Therapeutics, CEO Robert Habib is on a mission to develop a “new class of medicines” using small activating mRNA technology.
And on Thursday, the London-based biotech announced it landed a modest £23 million (nearly $30 million) Series A to work with.
Habib’s father, the prominent Imperial College London professor Nagy Habib, co-founded MiNA in 2008.
The company’s lead mRNA program is designed to “switch genes on,” and potentially help treat what the CEO called “the undruggable”: difficult-to-treat diseases, starting with liver cancer.
“It’s sort of encouraging the cell’s own machinery to pay attention and to start producing a bit more of the gene that we are interested in.
And that gene obviously will produce more messenger RNA. And that messenger RNA will result in more protein. And if we’re targeting the right protein, we’re going to have a therapeutic benefit,” Habib said.
In 2017, Sosei put down about $45 million for 25.6% of MiNA’s equity. Peter Bains, Sosei CEO at the time, laid out a $534 million plan to acquire MiNA, which was putting its lead candidate in a Phase I/IIa trial for liver cancer.
Bains said the deal was another step in expanding Sosei’s global reach. About a year and a half later, Sosei passed on that option, but kept its stake.

MiNA’s lead program goes after CEBPA in the hopes of reducing immune suppression caused by immature myeloid cells to improve the efficacy of cancer therapies.
Its candidate, MTL-CEBPA, is being tested as a combination therapy with Bayer’s sorafenib in advanced liver cancer patients and Merck’s pembrolizumab in patients with advanced solid tumors.
The drug doesn’t directly affect the cancer - just the environment in which it is growing.
“That strategy of reducing immune suppression and clearing those immature myeloid cells … is believed to be an important strategy in improving the effects of a range of cancer therapies,” Habib said.
The Series A funding will be used to conduct a Phase II study of MTL-CEBPA with sorafenib for the liver cancer indication, and complete an ongoing Phase I/Ib study of the pembrolizumab combination.
In the Phase II study, MiNA will focus on durability. Habib expects the study to begin early next year, with up to 70 patients in the UK, US, and some Asian and European countries.
When administered on its own, sorafenib typically has an objective response rate of between 0 to 10%, according to Habib. But when given with MTL-CEBPA,
“we’re seeing objective responses above that absolute proportion, and we’re seeing complete responses of liver patients which is an absolutely extraordinary feature,” he added.
“Liver cancer is one of the cancers which has really not improved by a huge amount over several decades,” Habib said. ” … The concept of complete remission is a concept which is very foreign to patients and to physicians in liver cancer.”
The company is actively looking to expand its platform to other indications as well.
aMoon, an Israel-based healthtech and life sciences venture fund, led the Series A round and existing investors pitched in.
As part of the deal, aMoon managing director Gur Roshwalb is joining MiNA’s board of directors.

MiNA Therapeutics has raised £23 million ($30 million) to advance a pipeline of small activating RNAs (saRNAs) aimed at previously undruggable targets.
The series A round will enable MiNA to move lead drug MTL-CEBPA into a phase 2 liver cancer trial in combination with Bayer’s Nexavar.
London-based MiNA has made a name for itself in recent years as a developer of saRNAs designed to drive the upregulation of target proteins.
The drugs are structurally similar to siRNAs but work in a different way once inside a cell.
Upon entering the nucleus, saRNAs recruit endogenous transcription complexes, leading to the production of new mRNA and, by extension, the targeted protein.
Boehringer Ingelheim teamed up with MiNA to apply the approach to nonalcoholic steatohepatitis and other fibrotic diseases in 2017.
Earlier this year, AstraZeneca began working with MiNA to assess the application of saRNAs to metabolic diseases.
MiNA has advanced its internal pipeline in parallel to those partnered programs, culminating in the delivery of phase 1b data on the use of MTL-CEBPA in combination with Nexavar earlier this year.
The readout encouraged MiNA to plan a phase 2 study, creating a need for cash to fund the work.
Israeli VC aMoon has met MiNA’s need for money, leading a £23 million series A with the support of existing investors in the biotech.

In aMoon, MiNA has gained the support of a VC that has supported companies including Adicet Bio,
a Regeneron-partnered gamma delta T-cell player that has received funding from groups including the investment wings of Novartis and Johnson & Johnson.
The money will fund a phase 2 designed to validate MTL-CEBPA, a drug intended to increase levels of the C/EBP-⍺ master regulator of myeloid cell differentiation.
Increasing levels of the protein could reduce the myeloid cell immunosuppression, thereby making other cancer drugs more potent.
The most recent evidence (PDF) to support the hypothesis comes from a phase 1b study of 36 patients with advanced hepatocellular carcinoma (HCC).
Among 16 viral HCC patients naïve to prior tyrosine kinase inhibitor treatment, there were two complete responses and two partial responses.
MiNA compared the data favorably to the results on Nexavar as a monotherapy, which achieved (PDF) a 1% rate of complete responses in a head-to-head comparison with Opdivo run by Bristol Myers Squibb.
MiNA is also evaluating whether MTL-CEBPA enhances the efficacy of checkpoint inhibitors such as Opdivo.
