When the King of Sweden presents Greg Winter with a Nobel chemistry prize for his antibody discoveries on Monday, he will be the 16th laureate from the UK Medical Research Council’s Laboratory of Molecular Biology in Cambridge.
During the past 60 years no other lab in the world has been more successful than the LMB in proportion to its size at picking up the greatest award in science.
Jan Löwe, who took over as director this year after working at the LMB for 22 years, is confident that its extraordinary scientific productivity, which has led some to call the lab a “Nobel factory”, will continue for decades to come.
“This lab has always worked best as a slightly maverick institution,” Mr Lowe said.
“Besides excellent science, we have stable core funding from the MRC. Our scientists have the freedom to work on long-term problems without having constantly to justify their research. Discoveries sometimes take decades to reveal their beauty and importance.”
Richard Henderson, who was LMB director from 1996 to 2006, came to the lab as a PhD student in 1966.
Last year he won a Nobel Prize for his part in the invention of cryo-electron microscopy, a new way of imaging biological molecules.
He is now working today to improve cryo-EM so that it becomes “the dominant technology for structural biology”. He says that collaborative interaction between staff is one reason for the lab’s success.
“We all go up to the canteen three times a day and enjoy suggestions from colleagues,” he said.
Sir Greg, this year’s LMB laureate, also praised the lab’s informality but said its funding model also helped researchers.
The lab receives a block grant worth about £40m a year from the government through the MRC, freeing scientists from relying on outside funding for individual projects.
“I have been at other places where everyone has a sense of ownership of their own equipment, which makes collaborative working hard,” he said, before setting off to the Nobel celebrations in Stockholm.
Future Nobel Prizes could come from a host of other projects at the lab. Neurobiology is an area that Mr Lowe is particularly keen to promote as director.
“I consider that now, for the first time, fundamental questions about how the brain works, such as the nature of memory, can be studied seriously through the methods of molecular biology,” he said.

When Greg Winter started his work, there was no way of knowing that it would lead to many new antibody-based drugs. Background:
Glutamatergic excitotoxicity is a recognised mechanism of neuronal injury and genetic and pharmacological evidence points to a role for mGlu5 in driving disease progression in the SOD1G93A mouse.
Existing mGlu5 antagonists, are hampered by poor pharmacology, limiting their potential for clinical translation. Heptares Therapeutics’ optimised mGlu5 NAM, HTL0014242, was designed using their proprietary STAR technology (1,2).
Objectives:
We set out to determine the pharmacology and therapeutic potential of HTL0014242 in SOD1G93A mice.
Methods:
We conducted pharmacokinetic (PK) and receptor occupancy (RO) studies of HTL0014242 directly in SOD1G93A mice to enable dose selection.
HTL0014242 was dosed orally, once daily, from 25 to 90 days of age at 3, 10 and 30 mg/kg to investigate effects on early disease progression in SOD1G93A transgenic mice.
A second study was conducted dosing at 30mg/kg from 25 or 75 days of age through to end-stage disease.
Motor neuron counts (Nissl) and measurement of microglial (IBA1) and astrocyte (GFAP) reactivity was determined in lumbar spinal cord sections.
Results: HTL0014242 (10mg/kg,po) showed good exposure in plasma and spinal cord (spinal cord/plasma ratio ¼5.4).
Treatment from 25 days of age at 3, 10 and 30mg/ kg showed marginal effects on motor function up to 90 days.
However, quantification of GFAP staining in lumbar ventral horn, indicated a profound dose dependent reduction in astrocyte activation (54%, 71% and 88% at 3, 10 and 30mg/kg (p ¼ 0.0039, one-way ANOVA).
IBA1 staining showed a similar trend (45%, 39% and 64% reduction at 3, 10 and 30mg/kg). This reduction correlated very well with the degree of receptor occupancy of mGlu5 by HTL0014242 in the same mice.
In the second study, the reduction in astrocyte activation was confirmed when mice were dosed from 25 (p50.0001) or 75 days of age (p50.0001) at 30mg/kg, and the reduction in microglial IBA-1 staining was also recapitulated (25 days, p ¼ 0.0005, 75 days p ¼ 0.002).
However, a delay in onset of visible signs slowing of late stage disease progression and reduction in motor neuron loss only occurred in the group dosed from 75 days of age.
These mice showed a three-week extension in the time taken to fail on the rotarod test (p50.0001).
Discussion and conclusions:
These data suggest a key role for mGlu5 in driving glial, particularly astrocyte, activation.
The therapeutic benefit from dosing at an intermediate stage of disease (75 days) versus an early stage (25 days of age) ties with substantial evidence in the literature

Background:
The cholinergic neurons of the basal forebrain and medial septum provide the major source of cholinergic innervation to the neocortex and hippocampus and play a critical role modulating cognitive processes such as attention, learning and memory, in part through activation of post-synaptic M1 receptors.
It is widely accepted that Alzheimer’s disease (AD) is associated with significant early and progressive loss of cholinergic neurons.
Cholinesterase inhibitors including donepezil have modest efficacy, potentially because they target degenerating pre-synaptic cholinergic neurons.
An alternative and potentially more effective strategy is to target post-synaptic M1 receptors which are relatively preserved in AD.
Muscarinic receptor agonists including the M1/M4 agonist xanomeline and the M1 orthosteric agonist GSK1034702 have shown promising early clinical effects but were not further developed due to gastrointestinal and cardiovascular adverse events (AEs).
HTL0018318 is a selective M1 agonist currently under development for the symptomatic treatment of cognitive impairment in dementias including AD and Lewy Body Dementia (LBD).
The objective of this study was to examine the single and multiple dose safety, tolerability, pharmacokinetics of HTL0018318 in healthy younger adult and elderly subjects.
Pharmacodynamic biomarkers were also assessed but are not presented here.
Methods: The single ascending dose (SAD) study was a single centre, randomized, double-blind, placebo-controlled, sequential, single ascending oral (solution) dose study.
HTL0018318 was administered in 5 cohorts of 8 healthy younger adult subjects in ascending dose levels of 1mg, 3mg, 9mg, 20mg and 35mg and elderly subjects in ascending dose levels of 9mg, 15mg, 23mg, 30mg and 35mg.
Effect of food and CSF pharmacokinetics were examined following the 20mg dose.
The multiple ascending dose (MAD) study was a single centre, randomised, double-blind, placebo-controlled, sequential, multiple ascending oral (solution) dose study.
HTL0018318 was administered in 3 cohorts of 12 healthy younger adult subjects dosed at 15mg/day, 20mg/day and 25mg/day once daily for 10 days, and 3 cohorts of 12 healthy elderly subjects dosed at 15mg/day, 20mg/day and 25mg/day once daily for 10 days.
One elderly cohort was dosed at 35mg using a titration regimen (5 days on 20mg/day and 10 days on 35mg/day). Results:
Pharmacokinetics of HTL0018318 were well-characterized in all subjects at all single doses. Exposure in terms of Cmax and AUC was dose-proportional.
Absorption was rapid with a typical Tmax of 1.0-1.5h post-dose and an apparent mean half-life of 12-16h.
HTL0018318 was found to distribute into CSF (CSF:plasma ratio≈30%).
There was no food effect on AUC or half-life of HTL0018318.
Single doses of HTL0018318 were associated with mild dose-related AEs (with low incidence) in both younger and elderly subjects.
The most frequently reported cholinergic AEs included hypersalivation, hyperhidrosis and increases in blood pressure, particularly following the 35mg dose (younger adult) and 23mg and 35mg doses (elderly).
In younger adult subjects, doses up to 20mg were not associated with changes in systolic and diastolic blood pressure and heart rate.
However, the 35mg dose was associated with an increase in mean systolic and diastolic blood pressure (up to 10mmHg) and mean heart rate (up to 9.8bpm).
In elderly subjects, significant increases in mean systolic and diastolic blood pressure (up to 11.9mmHg) and mean heart rate (up to 6.3bpm) were observed in the 15-35mg dose range, with no clear evidence of dose-dependency.
Pharmacokinetics of HTL0018318 were well-characterized in all subjects after multiple doses (Figure 1).
The Inter-individual variability in exposure was moderate (6-46 %CV covering Cmax and AUC0-24h).
HTL0018318 in repeated administration up to 35 mg/day for 10 days was generally well-tolerated, with mild AEs (with low incidence) and some evidence for dose-dependency.
The most frequently reported cholinergic AEs included hyperhidrosis, chills, cold sweat, headache, somnolence and nausea, particularly following the 25mg dose (young adult) and 25 and 35mg doses (elderly).
Repeated administration HTL0018318 over 10 days was associated with some small statistically significant increases in blood pressure on day 1 (up to 8.7mmHg) compared to placebo with a decline in this difference with continued dosing.
There were no consistent or clear dose-response relationships.
HTL0018318 caused small increases in mean heart rate (up to 10bpm), although these increases were in the context of overall decreases in mean heart rate (i.e. smaller decreases with HTL0018318 relative to the placebo decrease).
There were no clinically significant observations or changes in blood and urine laboratory values or abnormalities in the ECGs and Holter assessments following both single and multiple ascending doses up to 35mg. Conclusions:
HTL0018318 showed well characterised pharmacokinetics and following single and multiple doses over 10 days were generally well tolerated in the dose range studied.
The initial increase in blood pressure following single doses tended to decline with repeated dosing while increases in heart rate were small relative to baseline.
These findings provide encouraging safety and pharmacokinetic data in support of the development of HTL0018318 as a symptomatic treatment for cognitive impairment in Dementia.

・SITraNのHeledd Brown-WrightはHTL0014242について講演を行い、Sosei Heptaresが設計した新規mGlu5ネガティブアロステリックモジュレーター(拮抗薬)が、運動ニューロン病モデルマウスの疾患進行末期の運動機能低下を遅らせたことを明らかにした。
・この結果より、対照群マウスと比較して運動ニューロンの死滅が減少したことから明らかなように、HTL0014242が神経炎症(グリア細胞の活性化)を抑え、神経保護作用を起こさせることができることが示された。
