この度、プロテアーゼ活性化受容体2 (PAR2)に対し高い親和性と選択性を有するペプチド・アンタゴニストを同定することができましたのでお知らせいたします。
この度の同定は、構造的に安定な受容体の精製、提供を行うそ－せい社のStaRプラットフォームと
ペプチドリーム社独自の創薬開発プラットフォームシステム：PDPS（Peptide DiscoveryPlatform System)を組み合わせることにより可能になりました。
当社とペプチドリーム社は、2017年6月に戦略的コラボレーション契約を締結したばかりで、両社は共同で創薬・開発プログラムを実施し、そのコストを分担し、得られた成果物を共同所有するものです。
PAR2は、疼痛、がん、炎症性疾患における複数の適応で十分に検証済みのGPCR標的です。
PAR2は、酵素によって開裂した受容体の開裂部位が受容体自身のアゴニストとして作用することで活性化する特殊なGPCRであり、
独特の活性化機序のため、従来の創薬アプローチでは、新薬の開発においてPAR2アンタゴニストを同定することは極めて困難なものでした。
現在は、最も有望なリード化合物の臨床開発への進展を目指し、本ペプチドの特性分析と最適化をさらに行っております。

24日（木）の8時30分に「そーせいグループ株式会社との戦略的共同研究において PAR2標的に対して高い親和性と選択性を有するペプチド・アンタゴニストを同定」をリリースしました。
当社とそーせいグループ株式会社の子会社であるHeptares Therapeutics社は、
昨年６月に炎症性疾患の治療において重要な役割を担っているGタンパク質共役受容体（GPCR）を標的とする新規治療薬の研究開発・商業化を目的とした戦略的共同研究契約を締結しました。
創薬ターゲットとした炎症性疾患の治療において重要な役割を担っているGPCRが「プロテアーゼ活性化受容体2（PAR2）」です。
PAR2は、細胞膜を７回貫通している構造の受容体で、体内の酵素（トリプシン等）によって受容体の細胞外にある端に近い箇所が切れ、
その切れた先端領域が、同じ受容体の第2ループに結合することで活性化し、細胞内にシグナルを送るという独特の活性化機序をもつGPCRです。
そのため、PAR2に働いて細胞内へ炎症促進のシグナル伝達を阻害する物質（アンタゴニスト）を見い出すこと（同定すること）は極めて難しいとされていました。
しかし、今回、両社のプラットフォーム技術（StaRとPDPS）を組み合わせることで、強い結合力と高い特異性を有する新規ペプチド・アンタゴニストが同定できました。
しかもわずか2カ月未満で同定することができましたのでニュースリリースとさせていただきました。

Abstract:
Glycopyrronium is a once-daily, inhaled long-acting muscarinic antagonist (LAMA) demonstrating similar efficacy to inhaled tiotropium in patients with moderate-to-severe COPD;
owever, the benefit of LAMAs on COPD symptoms has been variable.
COPD is a progressive disease in which many patients develop an acute or sustained deterioration.
Data on the prevention of clinically important deteriorations (CID) using LAMAs are limited.
A pooled analysis was performed on four Phase III trials (n = 2936) that compared the efficacy of glycopyrronium (n = 1859) with tiotropium (n = 1077).
The primary endpoint was significant delay and/or reduction in the occurrence of CID.
CID was defined as any of the following: ≥100 mL decrease from baseline in pre-dose forced expiratory volume in 1 second (FEV1), ≥4 point increase in St George’s Respiratory Questionnaire score
or a moderate-to-severe COPD exacerbation occurring after the first dose of study medication.
A sustained CID was a CID occurring on ≥2 consecutive visits 4 weeks apart or for ≥50% of all available subsequent visits.
Baseline characteristics for the overall population were similar. Patients had moderate (62%) or severe (38%) COPD. Mean post-bronchodilator FEV1 was approximately 55% predicted,
and mean FEV1 reversibility was 16.7 and 18.6% in the glycopyrronium and tiotropium groups, respectively.
Both glycopyrronium and tiotropium significantly reduced time to CID and sustained CID versus placebo (p < 0.001).
No statistically significant differences were found between the glycopyrronium and tiotropium treatment groups in time to CID or sustained CID.
Glycopyrronium is effective in delaying time to clinically important deteriorations, with similar efficacy to tiotropium.

・Dr Chris Tate is a Programme Leader at the MRC Laboratory of Molecular Biology, Cambridge, where he researches G protein-coupled receptors.
・He is a co-founder of Heptares Therapeutics, a company that is creating novel medicines targeting G protein-coupled receptors.
・Recently, the lab of Dr Tate has engineered a minimal G protein, mini-Gs, that improves the chances of getting a high-resolution structure of a receptor in the fully active state.

[Chris Tate: "Cryo-EM of GPCRs"]: "Structural StudiesStructure of activated states of G protein-coupled receptors by Electron Cryo-Microscopy."
・The structures of G protein-coupled receptors in multiple different conformations are necessary to understand how agonists activate the receptor to facilitate G protein coupling.
・In addition, structures are also required bound to arrestins to understand the molecular mechanism of biased agonism.
・We have developed a thermostabilisation technology that has allowed us to determine the structures of multiple different GPCRs bound to inverse agonists, partial agonists, biased agonists and full agonists.
・Recently, we also developed a thermostabilised G protein that has allowed us to determine the structure of the activated state of the adenosine A2A receptor.
・We wish to build on these successes and to utilise the developments in electron cryo-microscopy to determine structures of GPCRs bound to stabilised heterotrimeric G proteins and arrestin by single particle imaging.
・The successful candidate will be expected to learn all the steps involved in the overexpression, purification and preparation of cryo-EM samples of a GPCR
・and to determine one or more structures using the Titan Krios equipped with state of the art detectors.

As the Royal party entered the spacious central atrium they got their first glimpse of the interior of the magnificent building, which was completed to budget and only a few weeks behind schedule.
Costing £212 million, the new building provides world-class research facilities and space for around 600 scientists, PhD students and support staff.
The new LMB building is the flagship for the extended Cambridge Biomedical Campus and the official opening is a highlight of the Medical Research Council’s Centenary celebrations, taking place throughout 2013.
The Queen viewed a display of items from the LMB Archive that showed a sample of the work that has led to nine Nobel prizes for research undertaken at the LMB.
This was followed by a presentation on how the LMB has exploited its discoveries - through patents, licensing and business start-ups –
which has helped to advance medical research and improve the UK’s economic competitiveness.
The Queen spoke at length to LMB group leader, Chris Tate, and was fascinated by the potential for new drug delivery systems.
Meanwhile, The Duke of Edinburgh was shown a timeline of the lab’s successes.
This included a presentation about the pioneering research in the area of structural studies - the determination of the 3D structure of biologically important molecules, such as proteins.
