There has been a lot in the press during the past year that pertains to Alzheimer’s disease.

Allergen and Heptares:
Despite setbacks, continuous effort has been leveled at amyloid and Tau.
There is still another aspiring player that is attempting to take a different approach.
Ireland’s Allergan (AGN) is using a notable proportion of its resources toward Alzheimer’s.
As of November 22, it has announced the acquisition of Chase Pharmaceuticals which has a late stage product candidate that is intended to improve dosage delivery of the current standard of care.
The purchase price of $125 million seems low for a viable product.
Of course, any success would be wonderful.
Perhaps even more interesting about Allergen’s research is its recent partnership with Heptares, based in the United Kingdom, but owned by a Japanese entity.
A Heptares founder is actually a 2017 Nobel laureate in chemistry.
Heptares specializes in G Protein-Coupled Receptors (GPCR) which can present promising, but elusive, drug targets for a wide range of conditions.
Brain receptors known as muscarinic M1 and M4 belong to a family or subfamily of GPCRs.
The sometimes-despondent healthcare blogger Derek Lowe has an informative write-up that describes past investigation into muscarinic M1, which is part of what Allergen has bought into.
His piece discusses Allergen’s partnership and concludes that “Leaps of faith are all we’ve got in Alzheimer’s.”
It may provide context for the present.
Heptares own descriptions of its M1 technology can be found here1, here2 and here3.
The deal with Allergan also contemplates M4, which is interesting:
"…M4 selective compounds may provide a novel approach to treat the neurobehavioral symptoms (psychoses) associated with Alzheimer’s disease and related neurological disorders,
through a different mechanism of action than available antipsychotics.
Combined, dual M1/M4 agonists may be able to treat both cognitive impairment and neurobehavioral symptoms."
Heptares has recently begun Phase 1 dosing of its M4 agonist.

HAIFA, ISRAEL, October 16, 2017 -
Pluristem Therapeutics, Inc. today announced the publication of results from a preclinical study in the peer-reviewed journal STEM CELLS Translational Medicine.
The article, titled “Placenta-derived adherent stromal cells improve diabetes mellitus-associated left ventricular diastolic performance”,
highlights the ability of PLX cells to significantly improve cardiac function and describes the underlying mechanism of action.
Results showed that treatment with PLX cells lead to improved diastolic function by significantly decreasing cardiomyocyte stiffness, endothelial inflammation, and improving vascularization.
The study was performed by investigators from the Berlin-Brandenburg Center for Regenerative Therapies, (BCRT) and the Charité-Universitätsmedizin Berlin, Germany, led by Professor Carsten Tschöpe.
Dr. Tschöpe is also a member of the Translational Research Committee of the Heart Failure Association of the European Society of Cardiology.
In the study, diabetes mellitus was induced in immune competent mice by streptozotocin application during 5 subsequent days.
Seven days after the first streptozotocin injection, animals were intravenously (IV) treated with either PLX cells or saline (placebo).
Cardiac parameters were assessed two weeks later.
The treatment using PLX cells led to improved diastolic function as indicated
by the heart-rate independent 1.2-fold (p<0.005) lower time constant of LV relaxation parameter Tau and the 1.2-fold (p<0.05) increase of the relaxation parameter dP/dtmin.
“Currently, there are limited treatment options for diastolic dysfunction and even fewer for diabetesinduced diastolic dysfunction," said Dr. Tschöpe.
“This study holds promise that PLX cells could potentially treat cardiac damage in diabetic patients, particularly in early-stage diabetic cardiomyopathy.
PLX cells are particularly well suited for this indication because they can be used without the need for tissue matching or immunosuppression.”
“Diabetes-induced diastolic dysfunction is a chronic disease that represents a large unmet need.
In this study, PLX cells were able to improve cardiac function when administered by simple IV injection.
This opens a potentially new method for an effective, low risk treatment for diastolic dysfunction,”
said Zami Aberman, Chairman and Co-CEO of Pluristem.
“These new data, combined with findings published in the Journal of Surgical Research,
which showed that PLX cells were effective in treating cardiac ischemia, suggest that PLX cells have the potential to address a wide range of cardiac disorders.”

G-protein-coupled receptors (GPCR) have been targeted in drug discovery research for decades due to their diverse roles in physiological and pathophysiological settings.
Activation of GPCRs by endogenous agonists is tightly regulated by desensitisation and internalisation mechanisms to maintain control of receptor activation.
However, several examples of sustained signalling have recently emerged which appear to bypass these regulatory processes.
The mechanism for these wash-resistant responses can potentially include rebinding, membrane deposition, signalling from internalised compartments, exosite binding or residence time.
We’ve investigated the potential mechanisms for sustained signalling at the GPR119 and adenosine A2a receptors suggesting a role for binding kinetics.
These examples, and others, highlight the importance of understanding the temporal aspect of agonist response profi les in drug discovery.
Moreover, optimising agonist residence time may enable us to attain more selective response profi les or to bypass these regulatory mechanisms to specifi cally target sustained signalling.

In recent years, G protein-coupled receptor (GPCR) engineering has shown remarkable development.
Amongst varied approaches, the StaR technology has been paramount in solving issues relating to GPCR instability:
a small number of point mutations are introduced in the protein, conferring high thermostability and selecting a specific receptor conformation.
These developments have led to a dramatic increase in GPCR high resolution structures and to new opportunities in structure-based drug design.
Meanwhile, ligand biding kinetics and residence time (calculated as the inverse of the dissociation rate constant) have become topics of increased interest in drug discovery.
In this presentation, data on small molecule binding kinetics and thermodynamics will be presented along-side new crystallographic structures
that help us understand GPCR structure-kinetics relationships and ligand selectivity.

・Heptares社と共同で、創傷治癒に関与するGタンパク質共役型受容体の1種であるBLT2を標的とした新規創薬研究開発プログラムを本年10月より開始します。
研究内容について：
本共同研究では、順天堂大学で横溝教授研究グループが発見した生理活性脂質受容体BLT2の作働薬候補を、
Heptares社がStaRテクノロジーを用いて分子モデリングし、数百種類の候補化合物を選定します。
それらの活性を順天堂大学が測定し、in vitroとin vivoで皮膚創傷治癒に対する治療効果を判定します。
この共同研究を通して、現在は難治性である褥瘡や糖尿病性皮膚潰瘍に対する新規治療薬の開発が期待されます。
また、これらの開発は、ケラチノサイトに作用する初めての皮膚潰瘍治療薬の開発につながるのみならず、
Gタンパク質共役型受容体の立体構造を分子モデリングして薬剤を開発するという新しい創薬アプローチにつながる可能性を秘めています。

順天堂大学大学院医学研究科 生化学・細胞機能制御学の横溝岳彦教授と、眼科学の岩本怜助教、松田彰准教授らの研究グループは、
生理活性脂質12-HHT(*1)とその受容体BLT2(*2)を介した角膜上皮損傷の修復メカニズムの解明に成功しました。
白内障術後などに処方される非ステロイド性消炎鎮痛剤(NSAIDs(*3))を含む点眼薬によって12-HHT産生が抑制される結果、角膜上皮損傷の修復が遅れることを見いだしました。
さらに、BLT2作動薬が角膜上皮障害の修復を促進することを発見しました。
これらの結果は、難治性角膜上皮障害の新規治療法の開発につながる成果です。
本研究は、英科学雑誌Scientific Reports電子版（2017年10月16日）に発表されました。
本研究成果のポイント：
・非ステロイド性消炎鎮痛剤は12-HHT産生を抑制することで角膜上皮損傷の修復を遅延させる
・BLT2作動薬は角膜上皮細胞の移動を促進することで角膜上皮障害の修復を促進する
・難治性角膜上皮障害の新規治療法につながる研究成果
(*1) 生理活性脂質12-HHT：
12-ヒドロキシペプタデカトリエン酸の略。
血液凝固時に主として血小板から大量に産生される炭素数17の不飽和脂肪酸。
細胞膜に存在するアラキドン酸という脂肪酸から細胞内の酵素反応によって産生される。横溝教授らの研究によって、生物学的な活性がはじめて報告された。
(*2) BLT2：
この受容体は、2000年の発見の際、別の生理活性脂質であるロイコトリエンB4(LTB4)に対する低親和性受容体であることが分かり、
高親和性受容体であるBLT1と区別するため、BLT2と命名された。
その後2008年に、より親和性の高い生体内リガンドとして12-HHTが発見された。
(*3) NSAIDs：
非ステロイド性消炎鎮痛剤(NonSteroidal Anti Inflammatory Drugs)の略称。
アスピリン、イブプロフェン、ロキソプロフェンなど、シクロオキシゲナーゼを阻害しプロスタグランジン産生を抑制することで、解熱鎮痛作用を発揮する。
短時間で効果を発揮するが、消化管粘膜障害、血液凝固阻害、陣痛発来遅延などの副作用が知られている。

Teva Pharmaceutical Industries Ltd., today announced the submission of a Biologics License Application (BLA) to the U.S. Food and Drug Administration (FDA) for fremanezumab,
an anti-calcitonin gene-related peptide (anti-CGRP) monoclonal antibody for the preventive treatment of migraine.

The BLA includes data from the HALO clinical trial program, which enrolled more than 2,000 patients with episodic migraine (EM) and chronic migraine (CM),
evaluating both monthly and quarterly dose regimens of fremanezumab. Results from these trials were recently presented
at the Congress of the International Headache Society (IHC) in September and will be published in future peer-reviewed publications.
The most common adverse events reported in clinical trials include injection site induration, erythema, and pruritis.
About the HALO Clinical Research Program:
The Phase III HALO EM and CM studies were 16-week, multicenter, randomized, double-blind, placebo-controlled, parallel-group studies to compare the safety, tolerability,
and efficacy of four dose regimens of subcutaneous fremanezumab compared to placebo in adults with episodic and chronic migraine.

Fremanezumab is a fully-humanized monoclonal antibody targeting the CGRP ligand, a well-validated target in migraine.

Shares of Israeli generics giant Teva Pharmaceutical Industries edged up nearly 1%,
after the company revealed that it has submitted a Biologics License Application (BLA) to the US Food and Drug Administration for fremanezumab,

Migraine market set to grow to $8.7 billion by 2026
Sales of migraine products are expected to grow to $8.7 billion by 2026,
at a compound annual growth rate (CAGR) of 10.3% across the seven major markets (5EU (France, Germany, Italy, Spain, and the UK), and Japan; 7MM),
according to business information and analytics firm GlobalData,
which adds that the USA will continue to dominate the market with a 77% share of total sales by 2026 followed by Germany (5.6%) and Italy (5.2%).
Other CGRP MAbs under development for migraine include erenumab, galcanezumab and eptinezumab, from Amgen, Eli Lilly and Alder BioPharmaceuticals, respectively.

Of the approximately 40% of patients suffering from migraine for whom prevention is appropriate, only 13% are currently receiving therapy.
There remains a significant medical need for treatments designed specifically to prevent migraine.
According to recent analysis, the economic burden for migraine patients reaches approximately $78 billion per year in the USA.

概要
国立大学法人大阪大学（以下、大阪大学）は、認知症性疾患を中心とした大脳疾患に対する専門医療およびそれに対する高度な治験を含む臨床研究を推進するために、
そーせいグループの100%子会社であるHeptares Therapeutics（英国、ヘプタレス・セラピューティクス、以下「ヘプタレス社」）の支援を得て、
10月1日に大阪大学大学院連合小児発達学研究科に行動神経学・神経精神医学 寄附講座を設置いたしました。
また本講座は、人材育成という目的に賛同いただいた協和会ならびに錦秀会からも支援を得ています。
現在、アルツハイマー病やレビー小体型認知症に対する薬剤の開発が世界中で進められ、多くの治験が進行中であり、さらに多くの治験が計画されています。
認知症性疾患や他の神経精神疾患による認知行動障害を対象とした治験においては、ヒトの大脳と認知機能との関係に関する知識、すなわち神経心理学の知識が必須とされますが、
我が国においてはそれに精通した専門医は少なく、また治験を遂行できるような神経心理学者はほとんど存在していないのが実情です。
過去のアルツハイマー病の共同研究の不成功の一因には、関与した研究者（医師）および心理士の技量の不均質性が指摘されていました。
また高度な内容の認知症をはじめとする神経精神疾患の治験を実施できる施設が極めて乏しく、原因疾患や重症度に応じた評価尺度を選択ないし開発できる専門家もごく少数であることから、
グローバルな治験への参加も困難で、我が国発の薬剤でも海外での治験を先行させるケースも多くみられました。
大阪大学大学院医学系研究科では、これまでも認知症の診療と臨床研究に力を入れ、新薬の開発にも大きく関わってきました。
一方、ヘプタレス社は、英国のMRC分子生物学研究所および国立医学研究所の研究成果を基に2007年に設立された医薬品開発企業で、
G-タンパク質共役受容体を標的とした創薬を行っています。アルツハイマー病およびその他認知症の治療薬の研究開発を行っています。
そこでこの度、これまで認知症診療や臨床研究の実績のある大阪大学において、本寄附講座を設け、認知症に対する高度な専門医療を実践するとともに、
認知症に対する薬剤の臨床薬理学の教育や研究の基盤を提供する場、すなわち「認知症医療イニシアティブ」を医学系研究科と協力して構築していきます。

Pluristem’s ARS program is developed and funded by the U.S. National Institutes of Health and the U.S. Department of Defense, and is in preparation for a pivotal study
HAIFA, Israel, Oct. 19, 2017 -- Pluristem Therapeutics Inc.,
a leading developer of placenta-based cell therapy products, announced today that the U.S. Food and Drug Administration (FDA)
has granted the company an orphan drug designation for its PLX-R18 cell therapy for the prevention and treatment of acute radiation syndrome (ARS).
ARS results from exposure to high levels of radiation, such as in the case of a nuclear accident or attack, and can lead to severe health consequences including death.
The Orphan Drug Act provides for granting special status to a drug or biological product, to treat a rare disease or condition.
The benefits of achieving Orphan Drug Designation include close guidance by the FDA,
which may accelerate the path to potential marketing approval, orphan drug grants, tax credits, and 7-year market exclusivity upon marketing approval.
Pluristem recently reported positive data from non-human primates (NHPs) studies of PLX-R18 cells
as a treatment for ARS conducted by the National Institute of Allergy and Infectious Diseases (NIAID) at the National Institutes of Health (NIH), U.S. Department of Health and Human Services (DHHS).
The reported data demonstrated improvement in survival rates of such NHPs and the enhancement of recovery across all three major blood lineages—white blood cells, red blood cells, and platelets.
In addition, PLX-R18 cells are also being studied by the U.S. Department of Defense’s (DOD) Armed Forces Radiobiology Research Institute (AFRRI)
to examine the effectiveness of the cells as a treatment for ARS prior to, and within the first 24 hours of exposure to radiation.
Pluristem PLX-R18 cells are also being studied by Fukushima Medical University in Japan for the treatment of ARS and as an adjunct to radiotherapy in cancer patients.
“Pluristem has a vast and dynamic program developing our PLX-R18 therapy as a treatment for ARS, which can potentially save many lives,”
said Zami Aberman, Co-CEO and Chairman of Pluristem.

is now available at pharmacies in the United States for the long-term maintenance treatment of airflow obstruction in people with chronic obstructive pulmonary disease (COPD), including chronic bronchitis and/or emphysema.
SEEBRI NEOHALER is a long-acting muscarinic antagonist (LAMA).
Sunovion entered into an exclusive license agreement with Novartis for the U.S. commercialization rights to SEEBRI NEOHALER, as well as Utibron Neohaler and Arcapta Neohaler , on December 21, 2016.

In two multi-center, double-blind, placebo-controlled, parallel group randomized trials,
SEEBRI NEOHALER demonstrated statistically significant improvements in change from baseline in lung function over 12 hours (FEV1 AUC 0-12h) compared to placebo (p<0.001).1,2 Improvements in lung function
were also measured by forced expiratory volume in one second (FEV1) and forced vital capacity (FVC) within five and 15 minutes post-dose versus placebo on Day 1 and Week 12.
Results showed reduced use of rescue medication and improvements in health-related quality of life,
as measured by the St. George’s Respiratory Questionnaire (SGRQ) total score, which is a composite of patient-reported symptoms, activities and impact on daily living.
“COPD is a serious, progressive respiratory disease that should have a personalized approach to choosing a therapy and delivery method, and there remains a need for additional treatment options

has launched the twice-daily long acting muscarinic inhibitor (LAMA) Seebri Neohaler (glycopyrrolate 15.6mcg), in the US.

The royalties earned upon net sales of Seebri Neohaler are the same as those for Seebri Breezhaler.

In December 2016, we announced that we signed a binding term sheet with Sosei Corporate Venture Capital Ltd., or Sosei CVC, for the establishment of a new Japanese corporation, or NewCo,

The parties plan to establish NewCo in Japan, in which we will own 35% of the equity in return for our contribution of a perpetual license to commercialize PLX-PAD for CLI in Japan.
All proprietary rights related to PLX-PAD will be exclusively owned by us.
Sosei CVC’s investment fund, Sosei RMF1, together with additional Japanese investors, will raise and invest approximately $11 million, equivalent to approximately \1.3 billion, in return for ownership of 65% of NewCo.

In December 2015, we reached an agreement with Japan’s Pharmaceuticals and Medical Devices Agency on the design of the final trial needed to apply for conditional approval of PLX-PAD cells in the treatment of CLI.
The approval of the protocol for the 75-patient trial was part of a larger agreement on the development of PLX-PAD via Japan’s new accelerated regulatory pathway for regenerative medicine.

In April 2015, Japan’s Pharmaceuticals and Medical Devices Agency, or PMDA, approved our large-scale manufacturing methods and quality for PLX-PAD cells for use in clinical trials.
This approval is an important milestone for initiation of a Phase I/II study in CLI in Japan.
We plan to submit an application for conditional, time-limited approval for marketing of PLX-PAD for treatment of CLI through Japan’s Accelerated Pathway for Regenerative Medicine.
The new regulatory pathway could potentially significantly reduce time to market for cell therapies such as PLX-PAD cells.
Two additional consultation meetings with PMDA were held at the end of July 2015 to discuss the safety of PLX-PAD and the design of a proposed study in CLI patients to be conducted in Japan.
In August 2015, the PMDA granted safety clearance to PLX-PAD cells for use in clinical trials in Japan,
and in December 2015 we received clearance for the clinical protocol and agreed with the PMDA on the terms for conditional marketing approval.
As a next step, we plan to submit a clinical trial notification to the PMDA to enable the initiation of the planned Phase I/II study of PLX-PAD in CLI following the expected establishment of a new Japanese corporation with Sosei CVC.

インターネット上で「暴騰銘柄」などと虚偽の情報を流した行為が金融商品取引法違反
（虚偽告知など）に当たるとして、金融庁は今月、投資助言サイトの運営会社３社を
相次いで行政処分した。証券取引等監視委員会によると、３社は虚偽情報を有料
で投資家に提供し、うち２社は助言サイトを格付けするサイト側と必ず上位にランク
される契約を結んでいた。監視委はこうした不正が横行しているとみて調べている。

Patent significantly expands potential indications for PLX-R18 and grants Pluristem broad coverage in the treatment of damaged hematopoietic systems
HAIFA, ISRAEL, October 23, 2017-- Pluristem Therapeutics Inc., a leading developer of placenta-based cell therapy products,
announced today that the European Patent Office has issued the company a patent titled,
“Methods for Treating Radiation or Chemical Injury” for its PLX-R18 cell therapy.
Pluristem currently holds several patents worldwide to cover placental 3D-expanded cells
in the treatment of impaired hematopoietic systems when a bone marrow, cord blood, or peripheral blood transplant takes place.
This important patent expands Pluristem’s IP assets to include a new set of indications related to the bone marrow’s inability to produce blood cells,
such as acute radiation syndrome (ARS), autoimmune diseases such as aplastic anemia, genetic disorders, chemotherapy, radiation therapy, and side effects from other treatments.
Pluristem currently holds over 115 issued patents and 100 pending patent applications worldwide.
“PLX-R18 is covered by several patents in key markets for the treatment of bone marrow failure following a transplant of hematopoietic stem cells,
or for the support of such a transplant.
What makes this a key patent for our IP assets is that it addresses the treatment
of a much broader range of medical conditions in which bone marrow is damaged and the patient has not received a transplant,”

・The COPD portfolio, which consists of Ultibro Breezhaler, Onbrez Breezhaler and Seebri Breezhaler, declined -1% (cc) to USD 165 million.
All three products in the COPD portfolio are delivered via the low-resistance Breezhaler inhalation device.
In the US, Sunovion Pharmaceuticals Inc. has assumed as of December 21, 2016 US commercialization rights
for these US products under the names Utibron Neohaler, Arcapta Neohaler and Seebri Neohaler.
Novartis will continue to bring Ultibro Breezhaler, Onbrez Breezhaler and Seebri Breezhaler to patients with COPD outside of the US.

Vectura Group plc, an industry-leading device and formulation business for inhaled airways products, confirms that Novartis,
has today reported Q3 2017 net sales of $101 million for Ultibro Breezhaler and $37 million for Seebri Breezhaler.
Total reported net sales for Ultibro grew 6%, and were impacted by short term normalisation of stocking effects in certain territories outside Europe.
In Europe, which accounts for the majority of reported net sales, Ultibro grew 18% year-on-year.
Total Seebri net sales were in line with the comparative prior year quarter, as expected.
The Vectura Board confirms its continued growth expectations for Ultibro’s long term sales potential and outlook for recurring revenue generation for the Group.

Abstract:
Small activating RNAs (saRNAs) are short double-stranded oligonucleotides that selectively increase gene transcription.
Here, we describe the development of an saRNA that upregulates the transcription factor CCATT/enhancer binding protein alpha (CEBPA),
investigate its mode of action, and describe its development into a clinical candidate.
A bioinformatically directed nucleotide walk around the CEBPA gene identified an saRNA sequence that upregulates CEBPA mRNA 2.5-fold in human hepatocellular carcinoma cells.
A nuclear run-on assay confirmed that this upregulation is a transcriptionally driven process.
Mechanistic experiments demonstrate that Argonaute-2 (Ago2) is required for saRNA activity,
with the guide strand of the saRNA shown to be associated with Ago2 and localized at the CEBPA genomic locus using RNA chromatin immunoprecipitation (ChIP) assays.
The data support a sequence-specific on-target saRNA activity that leads to enhanced CEBPA mRNA transcription.
Chemical modifications were introduced in the saRNA duplex to prevent activation of the innate immunity.
This modified saRNA retains activation of CEBPA mRNA and downstream targets and inhibits growth of liver cancer cell lines in vitro.
This novel drug has been encapsulated in a liposomal formulation for liver delivery,
is currently in a phase I clinical trial for patients with liver cancer, and represents the first human study of an saRNA therapeutic.

Anti-CGRP mAbs have reached the regulatory submission stage of development, which means the long expected patent disputes can begin in earnest.
Teva won a battle in EU that was brought by Eli Lilly and Alder Biopharmaceuticals last year but has decided to go on the attack in the US,
where there are a lot of interested parties that will be watching the proceedings closely.
A lot of projected revenue is tied up in the handful of anti-CGRP mAbs that are now within months of approvals,
which makes the predictable quarrels over who holds the superior intellectual property (IP) a high stakes legal drama.

In addition to being highly relevant to Amgen/Novartis and Alder,
Bernstein analyst Ronny Gal noted that the lawsuit involving Teva and Eli Lilly will also be closely watched by companies that are developing small molecule CGRP inhibitors,

Heptares is a Drug Discovery Company working on G protein-coupled receptors (GPCRs) involved in CNS and metabolic diseases.
Our unique structure-based approach in discovering therapeutic agents for these diseases relies on a proprietary technology to stabilize GPCRs,
enabling their purification for structural and biophysical studies.
We are currently seeking an experienced protein biochemist to join our site in Zurich working towards GPCR structure determination to enable drug discovery efforts.
Our preferred candidate will have the following profile:
・PhD in biochemistry or related discipline;
・Experience in molecular biology;
・Experience in membrane protein expression methods in mammalian and insect cells;
・Experience in membrane protein purification using AKTA Pure systems;
・Experience in preparation of protein for membrane protein crystallization (lipidic cubic phase and vapor diffusion);
・Experience in setting up crystallization using automated systems would be an advantage;
・Experience in preparing protein and setting up LCP-FRAP of advantage;
・Ability to work flexibly within a team whilst being highly self-motivated and work independently;
・Analytical thinking and good problem solving skills;
・Excellent organizational and time management skills as well as the ability to work to strict deadlines;
・Fluency in oral and written English; a knowledge of German is preferred; and,
・Relevant lab experience would be preferred.
・The successful candidate will be expected to work independently, and the role will include the following responsibilities:
Cloning of plasmid constructs for expression of crystallization constructs;
・Expression of GPCRs in insect and mammalian cells;
・Characterization of GPCR expression in membrane and in solubilized form;
・Construct engineering to optimize GPCRs for crystallization;
・Purification of GPCRs and biochemical characterization using a range of analytical techniques such as size exclusion chromatography - develop and optimize processes;
・Progression of purified material into crystallization
・Careful documentation of experimental work; and,
・Work closely with molecular biologists, protein expression scientists, crystallographers and pharmacologists to help advance the project.

Abstract:
G protein-coupled receptors (GPCRs) are the most intensively studied drug targets, mostly due to their substantial involvement in human pathophysiology and their pharmacological tractability.
Here, we report an up-to-date analysis of all GPCR drugs and agents in clinical trials, which reveals current trends across molecule types,
drug targets and therapeutic indications, including showing that 475 drugs (~34% of all drugs approved by the US Food and Drug Administration (FDA)) act at 108 unique GPCRs.
Approximately 321 agents are currently in clinical trials, of which ~20% target 66 potentially novel GPCR targets without an approved drug,
and the number of biological drugs, allosteric modulators and biased agonists has increased.
The major disease indications for GPCR modulators show a shift towards diabetes, obesity and Alzheimer disease, although several central nervous system disorders are also highly represented.
The 224 (56%) non-olfactory GPCRs that have not yet been explored in clinical trials have broad untapped therapeutic potential, particularly in genetic and immune system disorders.
Finally, we provide an interactive online resource to analyse and infer trends in GPCR drug discovery.

By reducing Aβ production, γ-secretase inhibitors were considered able to treat Alzheimer’s disease (Aβ hypothesis).
In fact, nearly 50 clinical trials have been conducted using potential γ-secretase inhibitors for Alzheimer’s disease or several types of cancer.
However, all of these trials have failed, except for two studies which are currently ongoing.
Osaka University scientists found that some potential γ-secretase inhibitors such as semagacestat,
which have been used in large clinical trials that ended in failure, do not function as true inhibitors as originally expected,
but rather cause accumulation of toxic intraneuronal Aβ.
They proved this by introducing an original method to measure direct intracellular products of γ-secretase.
They commented that the application of their evaluation method may help develop truly effective drugs for Alzheimer’s disease (AD).
The study can be seen in Cell Reports and provides an explanation for why the clinical trials for Alzheimer’s disease drugs have failed
and gives new light on the discord between preclinical and clinical findings.
“Aβ accumulates in the brain at the very early stages of Alzheimer’s disease,”
explains Osaka University Associate Professor Masayasu Okochi, an expert on the disease who managed the project.
“Aβ generation is based on the activity of presenilin/γ-secretase which mediates the cellular production of Aβ.”
Of the promising sets of drugs for Alzheimer’s disease were γ-secretase inhibitors like semagacestat.
However, a clinical trial that began almost 10 years ago was terminated early because not only was semagacestat found to fail,
patient groups that received the drug showed exasperated symptoms compared to the placebo group.
This finding has put great doubt into the Aβ hypothesis.

Our tests suggest that molecularly targeted therapy should be thoroughly checked from all angles before its application to clinical studies.
The new function of γ-secretase suggested in this study needs further analysis,
which will contribute to the development of truly effective drugs for Alzheimer’s disease and several types of cancer,” he said.

ヘプタレスの研究者は最先端の科学を追究できて楽しそうだけど、株主はまだまだ辛抱の時。
今後も収益は厳しいから早くパイプラインで評価されるときが来るといいね。
